Flow Cytometry

Flow Cytometry is a critical laboratory skill, particularly for characterization and analysis in cell/gene therapy. In this course, you will learn advanced flow cytometry skills such as panel design, multi-color analysis, and advanced troubleshooting.

Core Learning Outcomes Upon successful completion of this course, students will be able to:

  • Understand the fundamental principles of flow cytometry.
  • Identify the components of a flow cytometer and describe their role.
  • Interpret instrument quality control data.
  • Understand how to determine what fluorochromes can be detected by a flow cytometer basedon fluorochrome spectra and instrument configuration.
  • Summarize characteristics of fluorochromes that are highly relevant in practice (single/tandem,brightness, fluorescence spillover and spreading potential).
  • Gain a working understanding of fluorescence compensation.
  • Plan a flow cytometry experiment using adequate controls.
  • Learn common first steps for troubleshooting.
  • Sample Preparation: Proper sample preparation is crucial for successful flow cytometry.
  • Training includes techniques for cell staining, fixation, and permeabilization.
  • Data Acquisition and Analysis: Participants learn how to acquire data using flowcytometers and analyze it using software tools. This includes gating strategies, population identification, and statistical analysis.
  • Troubleshooting and Maintenance: Flow cytometry training often covers troubleshooting common issues, maintaining instruments, and ensuring data quality.
  • Advanced Topics: Introduce student to other advanced topics like cell sorting, spectralflow cytometry, and high-dimensional analysis including small particles analysis such as virus, EV, exosome.

Overall, this flow cytometry training equips students with the skills needed to confidently use
flow cytometry in their work.
